To study the variant expressions of proteins caused by a high or low radiation dose, the effect of X-rays on the expression of proteins in cultured glial cells was examined by proteome analysis. When glial cells were exposed to a high dose of 5 Gy, the relative abundance of 2 protein spots changed on 2-D gels. On the other hand, when cells were exposed to a low dose of 0.1 Gy, the relative abundance of one protein spot, different from one induced by 5 Gy, changed on 2-D gels. These protein spots were identified by MALDI-TOF mass spectrometry after digestion by trypsin. One of 2 spots which were changed by 5 Gy was identified as peroxiredoxin 2, and it increased with time until at least 24 hours after irradiation. However, when radiation dose is low, it causes stimulative effects on cells like the radiation adaptive response for cell growth.
