Recently, we developed a new in vitro model system using goldfish scales. This system can be used to detect the activities of osteoblasts and osteoclasts with alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) as respective markers. Therefore, it can be used to analyze the co-relationship between osteoblasts and osteoclasts. To obtain basic data for a space experiment, effects of calcemic hormones, such as parathyroid hormone (PTH), calcitonin, vitamin D3, and insulin-like growth factor-I on osteoblasts and osteoclasts in the scale, were examined using this assay in the present study. PTH activated osteoblastic activity and then increased osteoclastic activity. Furthermore, we found that other calcemic hormones function in both osteoclasts and osteoblasts in the fish scale as they do in mammals. Thus, scale is a functional calcium reservoir and has a physiological significance role in the bone metabolism of teleosts.
