It has been shown that the endogenous levels of IAA that induces peg formation are significantly lower in the upper side of the transition zone than that in the lower side. To understand the mechanism of decrease of auxin in the upper side of the transition zone in response to gravity, cucumber cDNAs similar to PIN auxin efflux carrier gene were isolated and tissue specificity of their mRNA accumulation was investigated. The results indicate that mRNAs of CsPIN1, CsPIN4 and CsPIN6 accumulate in the transition zone of 24-h-old cucumber seedlings that initiate peg formation. CsPIN1 mRNA abundantly accumulates in xylem and endodermis. CsPIN4 mRNA abundantly accumulates in xylem. CsPIN6 mRNA abundantly accumulates in phloem and pith, and also accumulates in endodermis and cortex.
