The fulcrum is extracellular matrix (ECM) for a cell. Most sHSPs constitutively express in slow muscle cells and contribute to developmental reorganization of various cell systems as well as responding to environmental stresses. Gravity and mechanical stress may influence cell architecture, so the increase and/or decrease of mechanical stress may induce molecular chaperones to maintain dynamics of solid system cytoskeleton inside of the cell and ECM protein like collagen protein outside of the cell. Similar to the changes of alpha-B-crystallin in atrophied soleus muscle under hindlimb suspension, HSP47 decreased with HS and increased after 3-G centrifugation. The effect of gravity clearly appears in tonic type of tension development, observed in slow skeletal muscle cells and cardiac myocyte. Dynamic property is produced and maintained with tonic tension development, in which tension development is realized by two different phases of contractions and relaxation in striated muscles. Former is consisted of the interaction of myosin and actin filament, and probably latter microtubule extension. Alpha-B-crystallin may work as molecular chaperone for tubulin/microtubule dynamics, so dynamic images of GFP-alpha-B-crystallin transfected by adeno virus vector was examined by time-lapse video system. Striated patterns of GFP-alpha-B-crystallin were obtained in beating myocyte. This suggest that alpha-B-crystallin may associate with protein complexes around probably Z/I bands receiving mechanical stress induced through contraction/relaxation
