Seedlings grown in a horizontal position suppress the development of the peg on the upper side of the transition zone in response to gravity. To understand how the response to gravity suppresses peg formation, cucumber mRNAs were screened by fluorescent differential display (FDD) to identify the mRNA that accumulates more in the non-peg side than in the peg side. First, to detect RT-PCR products from cucumber RNA the condition of FDD method was improved. Then, CsGRP1 cDNA that encodes glycine-rich protein was isolated using the modified condition of FDD method. Northern blotting showed that patterns of CsGRP1 mRNA accumulation corresponded to the results of FDD; namely that CsGRP1 mRNA accumulated more in the non-peg side than that in the peg side of the transition zone when peg initiation occurs. However, gravitropically bending hypocotyls did not show such differential expression of CsGRP1.
