Alaska) epicotyls grown true microgravity conditions in space has shown to be reduced as compared to that in 1 g conditions. Simulated microgravity conditions on a three-dimensional clinostat and the application of inhibitor of auxin polar transport, 2,3,5-triiodobenzoic acid (TIBA), have also been effective to reduce the auxin polar transport. Strenuous effort to learn how gravity contributes to the auxin polar transport in etiolated pea epicotyls in molecular levels resulted in successful identification of putative PsPIN1 and PsAUX1 genes encoding efflux and influx carrier proteins of auxin located in plasma membrane, respectively. PsPIN1 gene was isolated using RT-PCR techniques and cloned by TA cloning method. Significantly high homology was found on nucleotide and deduced amino acid sequences among PsPIN1 and AtPIN1 to AtPIN7 genes. Expression of PsPIN1 and PsAUX1 genes in etiolated pea epicotyls observed by RT-PCR analysis was not affected by simulated microgravity conditions but promoted significantly in root tip of the seedlings. The application of PCIB (anti-auxin, p-chlorophenoxyisobutylic acid) suppressed expression of PsPIN1 and PsAUX1 genes but TIBA had almost no effect. Based on the data described above, a possible role of PsPIN1 and PsAUX1 for auxin polar transport in etiolated pea seedlings will be
