UOEH","bibliographic_titleLang":"en"}]}]},"item_7_description_16":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"アメリカ航空宇宙局による有人月面探査構想が発表され,我が国も参加を表明している.月面には,月レゴリスと呼ばれる微粒子を含む表土が堆積しており,月面での活動によって月レゴリスが舞い上がり,宇宙船内に持ち込まれて吸入する可能性がある. 月面探査に先立ち,月レゴリスの有害性評価が必要であることから,本研究では月レゴリスの化学組成と粒径に着目し,細胞に対する影響を検討した.アポロが持ち帰った月レゴリスと同様の組成を持つ月レゴリスシミュラントを用い,液相沈降法により粒径が10 μm以下,25 μm以下および10~25 μm となるように分画した.さらに,平均粒子径5.10 μmの微粒子からなる月レゴリスシミュラントを用い,0.1および1.0mg/ml の濃度でヒト肺胞上皮腺癌由来のA549細胞およびマクロファージ様に分化させたヒト単球由来のTHP-1細胞に投与し,24時間後に,細胞傷害,酸化ストレス誘導および炎症誘導性を評価した.濃度1.0 mg/ml のときには,細胞膜の損傷とミトコンドリア活性の低下と,インターロイキン- 8( IL-8)の分泌および遺伝子発現の上昇が認められたが,0.1 mg/mlではこれらの影響はないか,ごく小さく,陽性対照とした結晶性SiO2に比べても影響は小さかった.THP-1細胞において,濃度1.0 mg/mlでは培養上清中にIL-1βおよび腫瘍壊死因子-α( TNF-α)の分泌がみられたが,24時間の時点でIL-1βおよびTNF-α遺伝子の上昇はみられなかった.また,細胞内酸化ストレスの上昇も小さかった.細胞膜損傷,酸化ストレス誘発あるいは炎症誘発性といった細胞影響は< 10 μmの画分で強い傾向があったが,顕著な違いはなかった.以上の結果から,in
