The near infra-red absortion spectra of proteins and some compounds containing one or two peptide bonds so far observed by us have been summarized and the following conclusions ragarding the configuration of the polypeptide chain and its structural units have been derived. (1) The peptide bonds in the polypeptide main chain are always in the trans form. (2) The structural units of the polypeptide chain are either in E-form, or in B-fonn. (3) The main chains of some α-proteins are in the BBB……configuration, while those of others may be inthe EBBEBB……configuration. (4) In compounds containing two peptide bonds whose molecules can exist either in E-form or in B-form ～ conspicuous temperature dependence of the equilibrium ratio of these two forms has been observed. (5) The entropy difference between these two forms calculated from this temperature dependence explains quite reasonably the magnitude of the entropy change of some proteins on denaturation. (6) The electronic structure of the free N-H bond in the molecule forming B-configuiation is considerably different from that forming E-configuration. |
