For this purpose, we isolated novel amino acid substitution mutants defective in formation of cortical microtubules (lefty & spr) in the Columbia background. The mutant defective in synthesis of membrane sterols (hmg) in the Columbia background was also produced by crossing the WS background mutant with wild type Columbia.
